摘要 目的用3H-TdR掺入法观察转化生长因子β1(TGF-β1)对人眼眶成纤维细胞增殖的影响,探讨增生性玻璃体视网膜病变(PVR)的可能发病机制。方法人眼眶成纤维细胞体外培养,用不同浓度TGF-β1(0.1、1.0、5.0、10.0、100.0μg/L)对细胞进行处理,3H-TdR掺入法测定细胞放射性强度。结果 0.1、1.0、5.0、10.0浓度的TGF-β1作用后活细胞数增加,细胞3H-TdR摄入量增加。100.0浓度的TGF-β1对人眼眶成纤维细胞作用相反。结论 TGF-β1对人眼眶成纤维细胞增殖有双相调节性,其不同作用的发挥依赖TGF-β1的浓度,TGF-β1的促成纤维细胞的增殖作用是PVR的诱发因素。 Objective To investigate the influence of transforming growth factorβ1 (TGF-β1) on human orbital fi- broblasts(OFs) proliferation by 3H-TdR absorption and explore the mechanism of proliferative vitreoretinopathy (PVR). Methods Human orbital fibroblasts were cultured in vitro with different concentrations of TGF-β2 ( 0. 1,1.0,5.0,10.0, 100.0 p.g/L ). Cell proliferation was determined by 3 H-TdR absorption. Results TGF-β1 with concentration of 0.1, 1.0, 5.0, 10.0 μg,/L could promote the proliferation of OFs, markedly increasing the number of living OFs cells and the value of 3H-TdR absorption compared with the control group, while 100μL TGF-β1 had a counter-effect. Conclusion TGF-β1 has concentration-dependant effects on OFs. Different concentrations of TGF-β1 may have inverse effects on fibroblast. The promotion of fibroblast proliferation indicates that TGF-β1 may play an important role in PVR.
