摘要
目的:研究免疫增强活性较好的酵母葡聚糖硫酸酯(SPS)的修饰条件。方法:通过L9(3)4正交设计对氯磺酸-吡啶法制备SPS的工艺进行优化,并以MTT法测定其对小鼠脾淋巴细胞体外增殖的促进作用。结果:最佳修饰条件为氯磺酸∶吡啶=1∶6(V/V),反应温度70℃,反应时间3h,硫酸基取代度可达1.092。在50~200μg.mL-1内,最佳工艺所得SPS对小鼠脾淋巴细胞生长有显著促进作用,平均增殖率可达89.85%。结论:优化方法制备的SPS对小鼠脾淋巴细胞生长具有显著的促进作用。
Objective: To optimize the modification conditions for the preparation of sulfated glucan from Saccharomyces cervisiae(SPS) with high immune enhancement activity.Methods: SPS was prepared using chlorosulfonic acid-pyridine method by L9(3)4 orthogonal design,immunomodulatory activity of SPS was examined by the lymphocytes experiment.Results: The optimized conditions were volume ratio of chlorosulfonic acid to pyridine at 1:6,reaction temperature at 70℃ and reaction time for 3 h.The content of sulfur in SPS and the degree of substitution(Ds) were achieved to be 12.78% and 1.09.In the range of 50~200 μg·mL-1,the lymphocytes-activating activity was significantly enhanced by SPS compared with control group(P0.01),with the mean proliferation rate at 89.85%.Conclusion: The SPS prepared by the proposed method has higher sulfur content and enhances the proliferation rate of lymphocytes from the spleen of ICR mice significantly
