红鲫DMRT1基因保守区的克隆与序列分析

摘要 目的进一步理解鱼Dmrt1基因的进化。方法利用兼并PCR技术,从红鲫基因组中克隆和测Dmrt1基因保守区的3个序列,分别命名为RcDmrt1a-1、Rc Dmrt1a-2和RcDmrt1b。结果 RcDmrt1a-1与RcDmrt1a-2、RcDmrt1b核苷酸同源性分别为92.2%、83.7%,RcDmrt1a-1与RcDmrt1a-2这2个序列的氨基酸组成完全相同。它们与RcDmrt1b的氨基酸序列有4个碱基不同。结论红鲫Dmrt1基因与斑马鱼、中国鳄、鸡、人的Dmrt1基因相比,显示了红鲫Dmrt1基因在结构上的保守性,暗示了在进化中功能的保守性。 Objective To further understand the evolution Dmrt1 gene in fish.Methods Using degenerate PCR,three nucleotide sequences of conserved region of Dmrt1 gene were cloned and sequenced from genomic DNA in the red crucian carp.These sequences were respectively named RcDmrt1a-1,RcDmrt1a-2 and RcDmrt1b.Results At the nucleotide level,RcDmrt1a-1 shared 92.2% and 83.7% identities with RcDmrt1a-2 and RcDmrt1b respectively.RcDmrt1a-1 showed 100% homology with RcDmrt1a-2 at the amino acid level.There are four amino acid differences between RcDmrt1a-1,RcDmrt1a-2 and RcDmrt1b.Conclusion Compared with Dmrt1 genes from the zebrafish,Chinese alligator,chicken and human,Dmrt1 gene in the red crucian carp is conserved in the structure,suggesting that the red crucian carp Dmrt1 gene is conserved in the function during the evolution.
