腺苷心肌保护液对离体大鼠心肌能量代谢及离子含量的影响

摘要 为观察腺苷心肌保护液对大鼠心肌ATP及离子含量的影响, 采用离体大鼠工作心脏缺氧停搏120 m in, 采用高钾、高钾+ 腺苷及腺苷停搏液进行心肌保护。发现停搏末含腺苷的两组ATP含量明显高于高钾组, 且复灌后进一步恢复; 各离子含量均能保持平稳。高钾组复灌后细胞内Na+ 、Ca2+ 明显增高伴Mg2+ 的减少。结果表明: 腺苷心肌保护液能够改善心肌能量代谢, 维持停搏期间及复灌后细胞内离子含量的稳定, The effects of adenosine (Ado) cardioplegia on m yocardial energy m etabolism and intracellular ion hom eostasis w as studied.The ratisolated w orking hearts perfused w ith K- H buffer w ere subjected to 120 m in ofhypotherm ic (15℃) cardioplegic arrestinduced by either St.Thom as'cardioplegia II(STS, group C), STSplus Ado 1 m m ol/L(group AK) or m odified STS(K+ 5.9 m m ol/L) plus Ado 1 m m ol/L (group ANK) .At the end ofarrest, ATPlevels of m yocardium w ere higher in groups AK and ANK than in group C.The concentration of intracellular Na+ , K+ , Ca2+ , Mg2+ had no change in groups AK and ANK, w hile Na+ , Ca2+ w ere increased and Mg2+ decreased in group C.In groups AK and ANK, the ATPlevels during reperfusion recovered to near baseline values but tended to be further depress in group C.These results suggest that Ado cardioplegia im proves the m yocardial energy m etabolism and m aintains intracelluar ion hom eostasis.
