针刺对小鼠腹腔巨噬细胞的热休克蛋白、诱导性一氧化氮合酶及其基因表达的效应 被引量:8

ACUPUNCTURE EFFECTS ON INDUCIBLE NITRIC OXIDE SYNTHASE mRNA, iNOS PRODUCT AND HEAT SHOCK PROTEIN IN PERITONEAL MACROPHAGES OF THE MOUSE

摘要 目的研究针刺对小鼠腹腔巨噬细胞的热休克蛋白(HSP)、诱导性一氧化氮合酶(iNOS)及其mR-NA表达的效应。方法将24只昆明小鼠腹腔注射无菌石蜡油后,随机分为3组:电针(EA)组、对照1(C1)组和对照2(C2)组。将3组收集的腹腔巨噬细胞制备成玻片和硝酸纤维素膜(NCM)两种标本,应用原位杂交、免疫细胞化学、细胞化学及斑点印迹技术检测HSP70、iNOS及iNOSmRNA。结果HSP70定位于巨噬细胞的胞质和胞核;iNOSmRNA及iNOS均定位于巨噬细胞的胞质;3组的iNOSmRNA、iNOS、HSP70的斑点印迹的扫描数值均为EA组>C2组>C1组(P<0.01)。结论电针可显著提高小鼠腹腔巨噬细胞的HSP70、iNOS及iNOSmR-NA表达。 Objective\ To study the acupuncture effects on the inducible nitric oxide synthase (iNOS) mRNA, iNOS product and heat shock protein(HSP) 70 in the mouse macrophages. Methods\ After peritoneal stimulation with steriled paraffin oil for 48h, 24 Kunming mice were randomly divided into 3 groups. They included:electroacupuncture(EA) group, treated with EA; Control 1(C1) group, peritoneal macrophages treated with culture medium for 18h; and control 2(C2) group, treated with neither EA nor culture The macrophages in 3 groups collected from respective peritoneal cavity were prepared into two kinds of specimens including slide and nitrocellulose membrane (NCM) specimens. The iNOS mRNA, iNOS and HSP70 were detected respectively with in situ hybridization, cytochemistry, immunohistochemistry and RNA and protein dot blots. Results\ The results showed that the signals of iNOS mRNA and iNOS product were localized in the macrophage cytoplasm; the immunoreactivity (IR) of HSP70 localized in both cytoplasm and nucleus. The dot blot signal scanning value of those in 3 groups in comparison all showed as follows: EA group>C2 group >C1 group, P<0 01 .Conclusion\ Acupuncture effect could up regulate expression of iNOS mRNA and promote iNOS activity and immunoreactivity of HSP70.
