摘要
目的在哮喘大鼠模型中探讨孟鲁司特(MK)与一氧化氮(NO)及诱导性一氧化氮合酶(iNOS)的关系。方法用卵清白蛋白作为致敏原制备哮喘大鼠模型,采用免疫组织化学技术、原位杂交技术等方法,观察哮喘动物模型中MK对NO及iNOS表达的调节。结果哮喘组肺组织中iNOS阳性表达积分(3.80±1.32)明显高于正常组(1.30±1.16)各用药组iNOS阳性表达明显低于哮喘组(P<0.01);哮喘组肺组织中iNOSmRNA表达积分(3.70±0.95)明显高于正常组(1.40±1.27),地塞米松和孟鲁司特显著抑制iNOSmRNA的表达(P<0.01)。结论与糖皮质激素相似,MK可以降低哮喘大鼠肺组织中iNOS的阳性表达率及iNOSmRNA的表达。
Objective To research the regulation of montelukast (MK) on inducible nitric oxide synthase(iNOS) expression in the lungs of murine model of asthma. Methods Animal model of asthmatic SD rats was developed by being sensitized and challenged with ov'albumin. By morphology examination, immunohistochemistry, in situ hybridization, we observed the regulation of MK on NO and iNOS expression in lung tissue. Results In asthmatic group, lung tissue showed stronger immunostaining for iNOS and increased expression of iNOS mRNA [ ( 3. 80±1.32 ), ( 3. 70±0. 95 ) ] than control group[ ( 1.30±1.16) ,( 1.40±1.27) ] ( P 〈0.01 ), in DX and MK group, there were down regulation of iNOS immunoreactivity and iNOS mRNA expression compared with asthmatic group( P 〈0.01 ). Conclusion Similar with glucocorticoids, expression of iNOS and iNOS mRNA were inhibited by MK.
