摘要
目的噁唑烷酮类抗菌药是一类新型化学令合成抗菌药,具有抑制多重耐药革兰阳性菌的作用,本文建立高效液相色谱法测定新型噁唑烷酮类药物YC-12的含量及检查有关物质。方法采用Diamonsil C18柱(250mm×4.6mm,5μm),磷酸盐缓冲溶液(1000mL水+2g磷酸二氢钾+3.5mL三乙胺+3mL磷酸)-乙腈-甲醇(42:30:28)为流动相;流速为1mL/min:检测波长254nm,柱温为25℃。结果主峰能与相邻杂质峰达基线分离,YC-12的浓度在10~200μg/mL范围内线性关系良好,相关系数为0.9994,最低检测限为0.5ng。结论该法简便、准确,灵敏度高,专属性强,可以用于YC-12的含量及有关物质的测定。
Objective Totally-synthetic oxazolidinones are a new class of antibacterial agents for the treatment of the infections caused by gram-positive bacteria including the multidrug-resistant pathogens. To develop an HPLC method for determination of new oxazolidinone derivatives YC-12 and its related substances. Methods The chromatographic separation of YC-12 was performed on a Diamonsil C18 column with a mobile phase consisting of phosphate buffered solution(1000mL H2O with 2.0g potassium dihydrogen phosphate, 3.5mL triethylamine and 3mL Phosphoric Acid)- acetonitrile-methanol(42:30:28, v/v/v) with a flow rate of 1.0mL/min. The detection was monitored at 254 nm, and the column temperature was maintained at 25℃. Results YC-12 was completely separated from its related substances. The calibration curve of YC-12 was linear in the range of 10-200μg/mL with the correlation coefficient of 0.9994. The limit of detection was 0.5ng. Conclusion The method is simple, rapid, sensitive and specific, and can be used for determination of YC-12 and its related substances.
