摘要
目的:建立高效液相色谱法同时测定金叶接骨木茎叶中齐墩果酸和熊果酸含量的方法。方法:采用Thermo ODS-2HYPERSIL(250 mm×4.6 mm,5μm);流动相为甲醇-0.4%磷酸水溶液(80%:20%);检测波长210 nm,流速0.8 mL/min,柱温25℃。结果:齐墩果酸的线性范围为12.17~243.36μg/mL(r=0.999 9),回收率为98.50%(RSD为3.03);熊果酸的线性范围为3.25~64.92μg(r=0.9998),回收率为99.49%(RSD为1.37)。金叶接骨木茎中齐墩果酸含量(mg/g)为1.485,熊果酸为0.348;叶中齐墩果酸为0.636,熊果酸为0.437。结论:本法操作简单,测定结果准确,可用于金叶接骨木中齐墩果酸与熊果酸的含量测定。
Objective:To establish a HPLC method for the determination of oleanolic acid and ursolic acid content in the stems and leaves of gold leaf elder.Methods:Thermo ODS-2HYPERSIL(250 mm×4.6 mm,5 μm) was used.Mobile phase was -0.4% methanol phosphoric acid(80%:20%),with 210 nm of detection wavelength,0.8 mL/min of flow rate,25℃ of column temperature.Results:The linear range of the oleanolic acid was 12.17~243.36 μg/mL(r=0.999 9),with 98.50%(RSD 3.03) of recovery rate.The linear range of the ursolic acid was 3.25~64.92 μg(r=0.9998),with 99.49%(RSD 1.37) of recovery rate.The oleanolic acid content in the gold leaf elder stems(mg/g) was 1.485,and the ursolic acid was 0.348;but the oleanolic acid in the leaves was 0.636,and the ursolic acid was 0.437.Conclusion:The method is simple and accurate and can be used for the determination of the oleanolic acid and ursolic acid content in the gold leaf elder.
