摘要
用冻干浓缩液的圆周免疫扩散法观察了牛胰岛素对 Hep G2 细胞分泌载脂蛋白apo AⅠ、apo AⅡ、apo B100、apo CⅢ及apo E的影响。结果发现,当培养液中的胰岛素浓度为1μg/m l时,apo AⅠ、apo E的分泌无明显改变,apo AⅡ的分泌有降低的趋势,apo CⅢ的分泌有增加的趋势,apo B100 的分泌降低21.2% ( P< 0.01); 当培养液中胰岛素的浓度为 10 μg/m l时,apo AⅠ的分泌减少6.9% ( P< 0.05),apo AⅡ的分泌减少 44.3% ( P< 001),apo B100 的分泌减少29.3% ( P< 0.01),而apo CⅢ的分泌增加51.8% ( P< 0.01),apo E的分泌无明显改变。该研究结果支持刘秉文提出的中国人内源性高甘油三酯血症发病机理的设想中的部分环节。
This study was intended to observe the effects of insulin on synthesis and secretion of apolipoproteins by hepatic cells.The HepG2 cells were cultured in RPMI 1640 media with bovine insulin (1μg/ml media and 10μg/ml media) for 72 hours, and the apolipoproteins contents in cultured media were measured by radioimmunodiffusion assay (RID) kits developed by authors' research laboratory. 40 fold lyophilizely condensed culture media were used for the assays. The results showed that the relative amounts of apo AⅠ, AⅡ, B100, CⅢ and E secreted by HepG2 cells detected by this method were 824±27, 813±24, 4875±82, 597±74 and 97±4 ng/mg cell protein, respectively. When 1.0μg/ml medium of bovine insulin was added into the culture media, apo AⅠ and apo E secretion remained unchanged.When compared with the control, while apo AⅡ tended to decrease, apo CⅢ tended to increase, and apo B100 secretion decreased by 21.2%; when 10.0μg/ml medium of insulin was added, apo AⅠ, AⅡ and B100 secretion decreased by 6.9% ( P <0.05), 44.3% ( P <0.01) and 29.3% ( P <0 01) respectively,while apo CⅢ secretion increased by 51.8%( P <0.01). These results support certain parts of the hypothesis of pathogenesis of Chinese endogenous hypertriglyceridemia proposed by Bing Wen Liu.
