摘要
目的通过对新疆塔里木马鹿鹿茸干鲜品为原料制备的多肽的提取方式、含量和在体外对小鼠前成骨细胞(MC3T3-E1细胞)增殖作用的比较,探讨新疆塔里木马鹿鹿茸的深加工和相关产品的开发原料选择的技术依据。方法通过酸性缓冲液抽提和超滤等方法制备得到马鹿鹿茸干鲜品多肽,以Folin-酚比色法测定其浓度,Tricine-SDS-PAGE电泳检测其相对分子质量(Mr)分布,MTT法检测其体外对MC3T3-E1细胞的增殖作用。结果马鹿鹿茸鲜品多肽主要由Mr为14 400,9 500,6 500和4 100的多肽组成;而马鹿鹿茸干品多肽在Mr14 400和4 100-6 500之间的多肽含量较低。体外活性分析结果显示,鲜品多肽能显著促进MC3T3-E1细胞增殖;而干品多肽促进MC3T3-E1细胞增殖效果不显著。结论马鹿鹿茸鲜品多肽具有更高的促进成骨细胞增殖的作用。
Purpose To compare the different contents of velvet antler polypeptides(VAPs) purified from the fresh and dry velvet antler of Cervus elaphus Linnaeus in Talim Xinjiang and their different effects on promoting the proliferation of mouse osteoblastic MC3T3-E1 cells in vitro,in order to provide evidence on selecting better raw material for deep processing and relative product design between the fresh and dry one.Methods VAPs were purified after several procedures,including acid buffer solution extraction,ultrafiltration,etc.The protein concentration of VAPs was determined by Folin-phenol method.Molecular weight distribution of VAPs was analyzed by Tricine-SDS-PAGE.Activity in vitro was analyzed by MTT assay promoting the proliferation of MC3T3-E1 cells.Results The molecular weight distribution of VAPs purified from the fresh velvet antler was mainly among 14.4,9.5,6.5 and 4.1 kD but that of the dry velvet antler was lack of 14.4 and 4.1-6.5 kD.The VAPs purified from the fresh velvet antler could highly promote the proliferation of MC3T3-E1 cells in vitro,but that from the dry velvet antler had no significant effect.Conclusion The VAPs purified from fresh velvet antler had higher bioactivity on promoting the proliferation of mouse osteoblastic cells in vitro than that from dry velvet antler.
