IL-29调节的肥大细胞膜表面和胞浆内蛋白酶激活受体表达分析

Modulation of IL-29 on membranous and cytoplasmic protease activated receptors expressed by P815 mast mells

摘要 目的:检测白细胞介素(interleukin,IL)-29引起的肥大细胞膜表面和胞浆内蛋白酶激活受体(protease activated recep-tor,PAR)-1,2,3,4表达。方法:肥大细胞P815培养后以不同浓度的IL-29激发肥大细胞,在不同时间点收集细胞,给予透膜和非透膜处理后用流式细胞术检测肥大细胞膜表面和胞浆内蛋白酶激活受体的表达。结果:IL-29激发肥大细胞2、6和16 h后肥大细胞膜表面和胞浆内PAR-1表达与相应对照相比差异有统计学意义,而IL-29调节的膜表面PAR-1表达与胞浆内PAR-1表达相比,差异无统计学意义。IL-29引起的肥大细胞膜表面和胞浆内PAR-2,3,4表达与相应对照相比,差异无统计学意义;而IL-29激发2 h肥大细胞膜表面PAR-2表达与胞浆内PAR-2表达相比,差异有统计学意义;IL-29激发6 h肥大细胞膜表面PAR-3表达与胞浆内PAR-3表达相比,差异有统计学意义;IL-29激发2、6和16h后肥大细胞膜表面PAR-4表达与胞浆内PAR-4表达相比,差异有统计学意义。结论:IL-29可以下调P815肥大细胞膜表面和胞浆内PAR-1表达,且IL-29引起的膜表面和胞浆内PAR-1表达无差异;尽管检测到PAR-2,3,4在膜表面和胞浆内的表达情况不同,但IL-29对膜表面和胞浆内PAR-2,3,4表达的调节作用不明显;流式细胞术检测的膜表面和胞浆内PARs表达结果均可反应IL-29调节的P815肥大细胞PARs表达情况。 Objective:To investigate the potential influence of IL-29 on the expressions of protease activated receptor(PAR)-1,2,3,and 4 on the membrane and in the cytoplasm of P815 mast cells.Methods:After being challenged with various concentrations of IL-29 for 2 h,6 h and 16 h,the P815 mast cells were collected and treated for flow cytometry to detect the membranous and cytoplasmic PARs expression.Results:Compared with the corresponding control,expression of PAR-1 induced by IL-29 was significantly decreased either on membrane or in cytoplasm of P815 cells,and the difference between membranous and cytoplasmic PAR-1 expression induced by IL-29 was not significant.IL-29 did little effect on the expressions of PAR-2,3,4 either on the membrane or in the cytoplasm of P815 cells as well as the corresponding control,though the differences between membranous and cytoplasmic PAR-2,3 and 4 were significantly detected.Conclusion:IL-29 can down-regulate the membranous and cytoplasmic PAR-1 expressed by P815 mast cells but has little effect on the expression of PAR-2,3 and 4 either on membrane or in cytoplasm of P815 cells.Though significant differences between membranous and cytoplasmic PAR-2,3 and 4 are present,either membranous or cytoplasmic PARs expression is representative while flow cytometry can be used to detect IL-29 modulating PARs expressed in P815 mast cells.
