摘要
为探讨中药水银(Hg)和轻粉(HgCl)的抗肿瘤作用,以三氧化二砷(AS2O3)及阿糖胞苷(Ara-c)为对照,应用细胞形态学、流式细胞术、MTT法和药物反应曲线等方法观察3种药物对K562和HEL细胞株的作用。结果表明,HgCl2和As2O3均能诱导562细胞凋亡,而Ara-c无此作用.HgCl2可使K(562)的G0/G1增高,提示激活P53基因可能是氯化汞诱导凋亡机制之一;HgCl2和AS2O3均可显著抑制K(562)增殖;Ara-c抑制HEL增殖作用明显强于HgCl2和As2O3三者对HEL均无诱导凋亡作用,但对K(562)和HEL均有不同程度杀伤和致死作用。HgCl2和As2O3作用相似,其主要机制可能是与Hg(2+)和AS(3+)与巯基(-SH)有极高亲和力,继而与含巯基的活性基结合、降低诸多酶的活性、损害细胞膜有关。HgCl2具有抗肿瘤作用,是一种新的诱导细胞凋亡药物。
To explore the effects of HgCl2 on K562 and HEL cell lines, and compared it with HgCl2 AS2O3 and Ara-c by using morphology, flow cytometry, MTT and drug-reactive curve. The results indicated that both HgCl2 and As2O3 could induce K562 cells apoptosis ,Ala-c did not have these effects; HgCl2 could increase percentage of K56, cells in G0/G1 phase. It indicated that HgCl2 induces K562 cells apoptosis probably by triggering P53 gene; As2O3 and HgCl2 inhibited the proliferation of K562 cells significantly; the proliferation of HEL cells was inhibited by Ara-c notably although it is not sensitive to As2O3 and HgCl2 .However, they all can not induce the apoptosis of HEL cell. HgCl2 is a new drug that can induce leukemia cells apoptosis.
