摘要
目的:探讨2型糖尿病(T2DM)患者外周血单个核细胞上果蝇样受体(TLRs)的表达以及1α,25-二羟维生素D3对TLRs表达的干预作用。方法:30例T2DM患者[其中伴有动脉粥样硬化(AS)者15例]和15名健康志愿者均抽取外周血,采用Ficol密度梯度离心法提取外周血单个核细胞(PBMCs),在不加或加入不同浓度1α,25-二羟维生素D3(10-8 mol/L、10-9 mol/L、10-10 mol/L)的情况下孵育48 h后,收集培养细胞,分别用流式细胞仪和定量RT-PCR检测细胞TLR2、TLR4表达水平。结果:与健康对照组相比,T2DM组PBMCs上TLR4水平增高,尤以伴有AS组明显,各组间TLR2表达差异无统计学意义。加入不同浓度1α,25-二羟维生素D3后,各组TLR4水平均有不同程度的下降,并呈现一定的剂量依赖性。结论:T2DM及其AS可能与TLR4水平增高有关;1α,25-二羟维生素D3能抑制体外培养的T2DM患者PBMCs TLR4的表达。
Objective: To probe the expression of TLRs of PBMCs and effects of 1α,25-hydroxyvitamin D3(1α,25-(OH)2D3)to TLRs on patients with Type 2 diabetes mellitus(T2DM)in vitro.Methods: the PBMCs from 30 patients with T2DM(15 of them had atheroscle-rosis(AS)) and 15 healthy controls were collected.Then,the PBMCs were treated with increasing concentrations of 1α,25-(OH)2D3.The expression of TLR2,TLR4 on PBMCs was detected by flow-cytometric analysis and RT-PCR.Results: The PBMCs of patients with T2DM showed higher expressions of TLR4 than those of controls,especially the patients with AS.But the expressions of TLR2 didn't show difference between T2DM and those of controls in our study.It showed that 1α,25-(OH)2D3 was found to dose-dependently suppress the protein and mRNA expressions of TLR4,but protein and mRNA expressions of TLR2 were no significance in vitro analysis.Conclusion: These results suggest that the inflammation triggered through TLR4 is important in the pathogenesis of atherosclerosis and T2DM.And it seems possible that 1α,25-(OH)2D3 may be used as a therapeutic option by modulating TLR4 expression of PBMCs in T2DM as well as complications.
