摘要
目的：探讨去甲二氢愈创木酸（Nordihydrognaiareticacid，NDGA）对恶性胶质瘤细胞裸鼠移植瘤的作用。方法：采用光镜和电镜观察移植瘤细胞的形态变化；用流式细胞仪进行细胞周期分析；用免疫组化检测肿瘤细胞中GFAP、PCNA的蛋白表达情况。结果：①NDGA可明显降低裸鼠移植瘤的成瘤率；②NDGA能抑制移植瘤细胞的生长和诱导其分化，使细胞增殖受阻于G1→S期。结论：NDGA在体内与体外有相同的抗胶质瘤作用。
Objective: To study the effects of nordihydroguaiarctic acid (NDGA) on the xenograft 0f human malignant glioma cells in nude mice. Methods: The morphological changes of xenograft cells were observed under optical and elecrron microscopy, cell cycle analysed by flow cytometry; and expression of GFAP and PCNA of xenograft cells were measured with immunohistochemistry in our study. ResuIts: ①NDGA inhibited the growth of the xenograft of human malignant glioma cell line SHG-44 in nude mice. ②NDGA induced the differentiation of the xenograft ceIls and blocked the cell growth cycle in G,-S phase. Conclusion:The therapeutic effects of NDGA on hurnan malignant glioma in vivo are similar to those in vitro.
