摘要
以盐酸小檗碱(BR)为荧光探针,研究了鲱鱼精DNA与橙皮素的相互作用。采用荧光光谱,紫外可见吸收光谱,盐效应,Scatchard方程等手段,探讨了橙皮素与DNA的作用机制.结果表明,在生理酸度(pH=7.4)下,橙皮素对DNA-BR体系的荧光有猝灭作用,猝灭类型为静态猝灭与动态猝灭共存的模式,其作用方式为嵌插和静电作用的混合模式,并求得其结合常数为1.83×104L.mol-1.
The interaction of Hesperidin with herring sperm DNA has been studied by fluorescence method using berberine (BR) as a fiuorescence probe. The mechanism of the interaction between Hesperidin and DNA was also researched by fluorescence spectrum, absorption spectrum, Scatchard equation, and salt effect. The results indicated that under the condition of physiological acidity (pH = 7.4), the fluorescence quenching model between HSP and DNA-BR belongs to a mixture of static fluorescence quenching and dynamic fluorescence quenching, HSP interacts with DNA by intercalation and electrostatic gravity. The binding constant calculated was1.83×10^4L·mol^1.
