摘要
酶法合成头孢立新的研究已有不少报道,并对酶的来源、测定方法、特性和应用等作了详细阐述。我们也作了大肠杆菌 AS1.76酶法合成头孢立新的研究,收率为86.9%,但侧链苯甘氨酸甲酯盐酸盐用量较大,且回收困难,阻碍了该法进一步推广。研究发现铜绿假单孢菌 AS1.204用以合成头孢立新可以解决这一问题。
From ten genera and 146 bacterial strains,22 strains producing α-amino acid ester hy-drolase were selected.Among them,AS 1.586and 41-2 were the best.The optimal conditions for synthesis ofcephalexin by pseudomonas aeruginosa 1.204were investigated.The optimal pH and tem-perature for enzymatic synthesis reaction waspH 6.8 and 25℃,respectively.By using1% 7-ADCA,3% PGME and 4% biomass,about 70% of 7-ADCA was converted to ce-phalexin under the mentioned conditions.
