摘要
从葡萄冠瘿中分离到的放射土壤杆菌 E26菌株,属于生物3型。在平皿培养时,能够产生土壤杆菌素,抑制葡萄根癌土壤杆菌的生长。在温室指示植物向日葵幼苗和葡萄幼枝上,明显抑制葡萄根癌土壤杆菌诱导形成冠瘿瘤。E26菌株对土壤杆菌菌株的抑菌谱较宽,其质粒的琼脂糖凝胶电泳图谱上,有两条质粒带,与生物3型葡萄根癌土壤杆菌 GL56-2菌株的质粒情况基本相同,其中大的质粒与根癌土壤杆菌 C58的质粒(PTiC58)大小近似。经双重抗药性标记的 E26菌株接种到葡萄枝条上,一个月后仍能得到抗药性菌株的分离,表明该菌株能在葡萄上定殖。
Crown gall on grapevines is a majorproblem in Northern China.On the basis ofsurvey of the distribution and incidence ofcrown gall disease on grapevines,a greatnumber of Agrobacterium strains were isolat-ed from tumor of grapevines.It was foundthat a grapevine isolate of A radiobacter,E26,belonging to nonpathogenic biotype 3produces an agrocin-like substance inhibitingpathogenic grapevine isolates of A.tumefa-ciens in vitro.In the greenhouse test E26also inhibited tumor formation on sunflowerseedlings and grapevine stems when strainE26 cells and pathogenic grapevine isolates ofA.tumefaciens were simultaneously inoculat-ed by equal number mixture.Strain E26produces agrocin with a broader spectrumof activity against grapevine isolates thanother A.radiobacter strains.Agrose gel elec-trophoresis showed that strain E26 harbor twoplasmids similar to the plasmids contained inpathogenic biotype 3 grapevine isolate G156-2.One of these,the bigger plasmid,had thesame relative mobility as the Ti plasmid fromstrain C58.The another plasmid was some-what smaller than the Ti plasmid.It is likelythat strain E26 harbors a mutant or deletiveTi plasmid and lose its virulence.Strain E26,which was made resistant torefampin and streptomycin,was able to co-lonize on the wounded site of grapevine stemafter it was inoculated on the stem for onemonth.
