摘要
以带小棒链霉菌bs1574为出发菌株,经γ射线照射,并结合甘油耐受性菌株的理性筛选,选育得到较佳诱变菌株bs2325,效价达2 569μg/mL。采用摇瓶发酵初筛和复筛,选育出甘油耐受性正向突变菌株,是出发菌株(效价为968μg/mL)的2.65倍,该突变菌株在琼脂斜面培养基上连续转接传代4代,克拉维酸的产量保持稳定。
The method of screening and selecting better mutants of bs2325 by γ-ray irradiation onto streptomyces bs1574 is put forward, and the titer is 2 569 ug/mL. By fermentation screening and secondary screening, strains of mutant glycerol positive tolerance is selected, whose titer is 2.65 times that of the original strain(968 ug/mL). The mutant strain grows well during four generations on the agar slant medium, and the yield of clavulanic acid remains stable.
