摘要
目的比较非活动性HBsAg携带者和健康人的血清蛋白标记物并建立相关筛选模型。方法用表面加强激光解析电离-飞行时间质谱技术(SELDI-TOF-MS)及CM10蛋白芯片对35例非活动性HBsAg携带者、35例健康人血清样本进行分析,将获得的血清蛋白质指纹图谱用计算机软件进行比较分析,建立非活动性HBsAg携带者的比较筛选模型。结果非活动性HBsAg携带者组与健康人对照组共有28个差异有统计学意义的蛋白峰;建立诊断模型的灵敏度94%(33/35),特异度80%(28/35),准确率为87%(61/70)。结论建立的血清蛋白质指纹图谱模型能够区分非活动性HBsAg携带者与健康人,SELDI技术在诊断及特异性血液生物标志分子的筛选等方面具有较好的临床应用价值。
【Objective】To screen relatively specific markers in serum from healthy people and non-active hepatitis B s' antigen carriers.【Methods】SELDI-TOF-MS Protein Chip was used to detect serum protein patterns of 35 non-active hepatitis B s antigen carriers and 35 healthy people,diagnostic model was developed and validated by using BPS.【Results】At the molecular weight range from 2000 to 50000,28 protein peaks were obviously differ-ent between non-active hepatitis B carriers and the healthy people.Analyzed by BPS,The diagnostic model separat-ed the non-active hepatitis B carriers from the control samples with accuracy of 87%(28/35),sensitivity of 94%(33/ 35)and specificity of 80%(28/35).【Conclusion】SELDI-TOF-MS Protein Chip array technology has better clinical application value in diagnosis and detection of specific serum biomarkers.
