摘要
目的:观察仙鹤草颗粒(herba agrim oniae granule, H A G) 对小鼠实验性糖尿病血糖的影响. 方法:采用链脲霉素(streptozotocin, S T Z)和肾上腺素(adrenaline, Adr)复制小鼠糖尿病模型,0.2,0.4,0.8 g/kg 仙鹤草颗粒ig,观察其给药不同时间的降血糖作用. 结果: ①0.8 g/kg H A G ig 12 d 有降血糖作用, 中、高剂量 H A G ig 8 d 和12 d 可明显降低 S T Z糖尿病小鼠血糖水平( P< 0.05~0.01),但 D860组给药4,8,12 d 均无明显降血糖作用. ② H A G中、高剂量组明显对抗 Adr 升高血糖作用( P< 0.05~0.01),并能显著增加动物肝糖元含量( P< 0.01),降低血糖水平( P< 0.01). ③中、高剂量 H A G 组能降低正常小鼠口服糖负荷后血糖的峰值( P< 0.05~0.01),并加快升高血糖水平回落的速度. 结论:该试药可对抗 S T Z和 Adr 引起的血糖升高,改善糖耐量,增加肝糖元合成;作用机制可能与其促进胰岛素分泌或增加组织对糖转化利用有关.
AIM: To explore the effect of Herba Agrimoniae Granule(HAG) on blood glucose in mice with experimental diabetes. METHODS: 0.2, 0.4, 0.8 g/kg HAG ig was given at different time spots to observe its effects of reducing blood glucose in STZ induced diabetic and adrenaline hyperglucemic mice. RESULTS: ①The serum glucose in normal mice was reduced when HAG with 0.8 g/kg was given for 12 d ( P< 0 05). HAG with 0.4 g/kg and 0.8 g/kg dosages could reduce blood and serum glucose in STZ induced diabetic mice after ig 8 d and 12 d respectively ( P< 0.05~0.01), but HAG administered to the positive group of D860 didn′t show significant blood glucose reducing effects. ②HAG with 0.4 g/kg and 0 8 g/kg showed a significant antagonistic effect on the blood glucose increased by Adr ( P< 0.05~0.01), significantly increased the liver glycogen and reduced serum glucose ( P< 0 01). ③HAG with midium and high dosages, after glucose was given, reduced the peak value of blood glucose ( P< 0.05~0 01) and sped up restoration of the increased blood glucose level. CONCLUSION: HAG can significantly check the STZ and Adr induced rise of blood glucose,improve glucose tolerance and increase hepatic glucogen synthesis. Authors of the paper suggest that this mechanism of reducing blood glucose is associated with HAG′s function of promoting regeneration of islet βcells or increasing tissue′ transformation and glucose utilization.
