HPLC法测定莪术油微乳液中莪术醇和吉马酮的含量 被引量:3

摘要 目的:建立测定莪术油微乳液中莪术醇及吉马酮含量的HPLC方法。方法:采用RP-HPLC法。色谱柱:ODS色谱柱;流动相:甲醇-水(80:20);流速:0.8 mL.min-1;柱温:室温;进样量:20μL;检测波长分别为:205nm(莪术醇)和245 nm(吉马酮);室温测定,莪术醇和吉马酮的保留时间分别为13.6和14.7分钟。结果:莪术醇和吉马醇的的线性范围分别为1.016~16.256mg.L-1和0.468~29.928mg.L-1;最低检测限分别为0.30μg/ml和0.021μg/ml,日内和日间变异系数小于10%,精密度大于90%。结论:该方法灵敏、快速、准确,操作简便、线性范围宽,可适用于测定莪术油微乳液中莪术醇和吉马酮的含量。 Objective:To develop a sensitive and specifie HPLC method for determination of curcumol and Germacrone in Zedoary Turmeric Oil emulsion. Methods: The mobile phase consisted of methanol - water ( 80 : 20, v/v). HPLC was run at the flow rate of 0. 8ml/min and were detected by UV absorption at 205nm and 24-5 nm, respectively. Temperature:room temperature. Retention time of Curcumol and Germacrone were 13.6 and 14. 7min, respectively. Results: Calibration plots were linear over the range of 1. 016 - 16. 256mg · L^-1 for Cureumol, and 0. 468 - 29. 928mg · L^-1 for Germaerone. The limit of quantitation was 0.30 μg/ml for Cureumol and 0. 021 μg/ml for Germaerone. The intra - and interday variations were less than 10% and aeeuraey was greater than 90%. Conclusion: The established method is proved to be sensitive, rapid and robust,and suitable for determination of Cureumol and Germaerone.
