扁平苔藓患者外周血CD4^+CD25^+ Treg细胞的检测 被引量:1

摘要 目的:检测扁平苔藓患者外周血CD4^+ CD25^+调节性T细胞(regulatory T cells,Treg)的数量及FOX3 mRNA的表达。方法:采用流式细胞术对25例扁平苔藓患者和25例健康对照者外周血CD4^+CD25^+ Treg细胞进行检测,并通过RT-PCR方法检测FOXP3转录因子在外周血单个核细胞的表达。结果:泛发性扁平苔藓和口腔扁平苔藓患者外周血CD4^+ CD25^+ Treg细胞数分别为(2.68±2.21)%和(2.38±2.67)%,均低于健康对照者(7.87±1.77)%;扁平苔藓患者的FOPX3表达水平也低于健康对照(P<0.05)。结论:CD4+CD25^+ Treg细胞数量和功能下降,可能通过导致扁平苔藓患者免疫功能紊乱参与了扁平苔藓的发病。 Objective: To investigate the number of CD4+ CD25+ regulatory T cells (Treg) in peripheral blood and the expression of FOXP3 transcriptional factor in the patients with lichen planus (LP). Methods: The amount ofCD4+ CD25+ Treg cells in peripheral blood was detected by flow cytometry and the expression of FOXP3 level in the mononuclear cells was detected by reverse transcriptase polymerase chain reaction in 25 patients with LP and 25 health controls. Results: The amount of CD4+ CD25+ Treg cells in the patients with generalized LP and the patients with oral LP was (2.68 ± 2.21) % and (2.38 ± 2.67) %, which was significantly lower than that in health controls (7.87 ± 1.77)% (P 〈0. 05). The expression of FOXP3 mRNA in CD4+ CD25+ Treg cells was also significantly decreased than that in controls (P 〈 0.05). Conclusion: Down- regulation of CD4+ CD25+ Treg ceils and FOXP3, leading to immunological dysfunction, may be involved in the pathogenesis of LP.
