摘要
本文利用壳聚糖为反应物制备硒化壳聚糖并对其硒含量检测方法进行了改进,考察了温度,搅拌时间,pH值及提纯方法对制备硒化壳聚糖的影响,得到最优反应条件为反应温度35℃,搅拌2 h,pH=5,用70%乙醇作为提纯试剂得到纯度较高,收率为78.6%的硒化壳聚糖。采用3,3’-二氨基联苯胺分光光度法来测定硒化壳聚糖中硒的含量,省去了高氯酸消化步骤,简化了实验操作,经验证对结果显著影响,可作为快速简便地测定有机硒含量的方法。
Chitosan was used for preparation of seleno-chitosan.The determination method tor the content of Se in seleno-chitosan was improved,and the temperature,stirring time, pH and purification method on the impact of seleno-chitosan preparation were inspected.The optimal reaction conditions were obtained:35 ℃,2 h,pH 5,with 70 % alcohol in the purification of high purity and the yield was 78.6 %. With 3,3'-diaminobenzidine spectrophotometry for determination of selenium content in seleno-ehitosan, the need for high chlorine acid digestion steps were eliminated, the experimental operation was simplified,there was no significant effect on the resuhs.The method could be used as the determination of quick and easy method to determine the content of organic selenium.
