摘要
利用SSR引物对44份外部形态差异较大的扁穗牛鞭草基因组进行扫描,结合农艺性状,进行性状与标记间的关联分析。结果表明,①扁穗牛鞭草群体的农艺性状存在较大变异,其中分蘖数变异最大,变异系数达34.8%,其次是单株生物量、叶长和茎叶比,变异系数也达20%以上。②扁穗牛鞭草农艺性状间存在显著的相关关系,单株生物量与叶宽、茎直径、茎长呈极显著正相关,茎叶比与节间长、茎长呈极显著正相关,与分蘖数呈极显著负相关。③通过单向分组资料的方差分析,从182条多态性条带中共检测到与扁穗牛鞭草8个农艺性状极显著相关的SSR标记18个,其中,6个标记分别与2个以上性状关联。标记对性状变异的贡献率为15.1%~32.8%,SG5-160能解释32.8%节间长表型变异,引物SG25的4个标记对茎直径表型变异的总贡献率达91.6%。该研究结果对早期发现优异种质,缩小筛选范围,降低筛选成本和工作量,加快扁穗牛鞭草育种进程有重要意义。
8 agronomic characters of 44 Hemarthria compressa materials were measured and 10 pairs of SSR premiers were used to amplify the genomic DNA of these materials.Single factor variance analysis was used to identify association between quantitive traits and SSR mocular markers.The result showed that: ① there were various variability among H.compressa germplasm.The variation coefficient of tillers was 34.8%.The variation coefficient of individual biomass,leaf width and stem-leaf ratio was more than 20%.②Significant correlation was found among these agronomic characters.There were significantly positive correlation between individual biomass and leaf width,stem diameter,branch length,and significantly positive correlation between stem-leaf ratio and internode length,branch length.However,the correlation between stem-leaf ratio and tillers was significantly negative.③The result of one-way ANOVA revealed that 18 markers out of the total 182 PCR produced bands were significantly associated with the 8 agronomic characters,among which 6 markers were associated with more than 2 traits.The contribution rate was from 15.1% to 32.8%,and the SG5-160 marker accounted for 32.8% of the phenotypic variation of internode length.The contribution rate of four markers of premier SG25 was account for 91.6% of the phenotypic variation of stem diameter.The result was useful to find excellent H.compressa germplasm early,reduce selection range,decrease screening cost and workload,and accelerate breeding process.
