摘要
从含油污泥中筛选出一株产脂肪酶活力较高的微球菌,此菌最适产酶条件:2%的蛋白胨为氮源,1%的橄榄油为碳源,pH8.0,28℃培养48h.酶学性质研究发现,在28℃和45℃脂肪酶有较高催化活力,其中28℃活力更高些,最佳反应pH为8.0,pH稳定范围在6.0~10.0之间,Ca2+和K+对酶有激活作用,Fe2+,Cu2+和Mn2+等对酶有抑制作用.
A mlcrococcus that produces lipase was isolated from soil by means of solid plate assay method. The optimal lipase-producing conditions of the strain were as follows: 1 % olive oil as carbon source, 2% peptone as nitrogen source, pH 8. 0, 28 C and the culture period of 48 h. The optimal pH and temperature for the hydrolysis of olive oil by partial purified lipase were pH 8. O and 28 C respectively. The enzyme;activity was stimulated by Ca2 and K+ but was inhibited by Ye2+, Cu2+ and Mn2+etc.
