摘要
从富油土壤中分离筛选到58株脂肪酶产生菌,其中GXL02菌株产脂肪酶的能力较强,根据其形态特征及16SrDNA序列分析,初步鉴定为不动杆菌。GXL02发酵产酶需油脂的诱导,同时也依赖Mg^2+,培养基中不添加Mg^2+时几乎不产酶,微量的Mg^2+就能激活菌体产酶。葡萄糖、蔗糖等糖类物质不利于GXL02产脂肪酶。该酶的最适作用温度为50℃,最适作用pH为9.0,60℃保温90min酶活基本不损失,在pH3.0~10.0范围内稳定。
58 strains producing lipase were isolated from rich oil soil. The lipase from GXL02 has the highest activity to catalyze glycerolysis of oil. Strain GXL02 was identified as Acinetobacter sp. according to its character and 16S rDNA sequence. Lipase production conditions of Acinetobacter sp. GXL02 were studied in detail. The results showed that lipase production of GXL02 depended on fat and Mg^2+. Glucose and some other saccharide carbon sources restrained lipase production. The optimal pH and temperature of the lipase for hydrolysis of olive were pH 9.0 and 50℃ respectively. Activity of lipase was no much change in 60℃ for 90 min, and stable from pH 3.0 to pH 10.0.
