摘要
目的研究鼻咽癌组织中Pokemon、Ku70的表达及其临床病理学意义。方法采用免疫组织化学Envision二步法检测Pokemon、Ku70在47例鼻咽癌(鼻咽癌组)、20例鼻息肉(鼻息肉组)石蜡包埋组织中的表达,探讨与TNM分期、浸润、转移的关系。结果 (1)Pokemon在鼻咽癌组及鼻息肉组中阳性表达率分别为68.1%(32/47)、25.0%(5/20),差异有统计学意义(χ2=10.53,P<0.05)。不同分型的鼻咽癌组织中,角化性鳞状细胞癌组与非角化癌组Pokemon的阳性表达率分别为45.0%、85.2%,差异有统计学意义(χ2=8.54,P<0.05)。不同TNM的鼻咽癌组织中,Ⅰ期+Ⅱ期组、Ⅲ期+Ⅳ期组Pokemon的阳性表达率分别为88.0%、45.5%,差异有统计学意义(χ2=9.75,P<0.05)。在有无淋巴结转移的鼻咽癌组织中,有淋巴结转移组、无淋巴结转移组Pokemon的阳性表达率分别为86.7%、35.5%,差异有统计学意义(χ2=13.18,P<0.05)。⑵Ku70在鼻咽癌组及鼻息肉组中阳性表达率分别为61.7%(29/47)、30.0%(6/20),差异有统计学意义(χ2=5.65,P<0.05)。不同分型的鼻咽癌组织中,角化性鳞状细胞癌组与非角化癌组Ku70的阳性表达率分别为55.0%、66.7%,差异无统计学意义(χ2=0.66,P>0.05)。不同TNM分期的鼻咽癌组织中,Ⅰ期+Ⅱ期组、Ⅲ期+Ⅳ期组Ku70的阳性表达率分别为72.0%、50.0%,差异无统计学意义(χ2=2.40,P>0.05)。在有无淋巴结转移的鼻咽癌组织中,有淋巴结转移组、无淋巴结转移组Ku70的阳性表达率分别为73.3%、35.5%,差异有统计学意义(χ2=4.75,P<0.05)。结论提示Pokemon及Ku70在鼻咽癌的发生发展中起重要作用;Pokemon与Ku70可能成为判定预后的生物标志物。
Objective To investigate the expression and significance of Pokemon,Ku70 in nasopharyngeal carcinoma.Methods Immunohistochemical Envision method was adopted to determine the expression of Pokemon and Ku70 in 47 cases of NPC and 20 cases of nasal polyp.Their correlations to the clinicopathologic characteristics of NPC were retrospectively analyzed.Results（1） Positive expression rate of Pokemon in NPC group and nasal polyp group was 68.1%（32/47） and 25.0%（5/20） respectively,with statistical significances（χ2=10.53,P〈0.05）.In different types of NPC issues,positive expression rate of Pokemon in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma was 45.0% and 85.2% respectively,and the expression difference was of statistical significance（χ2=8.54,P〈0.05）.In different TNM NPC tissues,Ⅰstage ＋Ⅱstage group,and Ⅲ stage ＋Ⅳstage group,positive expression rate of Pokemon was 88.0% and 45.5% respectively,and the difference was of statistic significance（χ2=9.75,P〈0.05）.The positive expression rate of Pokemon in NPC tissues with or without lymphatic metastasis was 86.7% and 35.5% respectively,and the difference was of statistic significance（χ2=13.18,P〈0.05）.（2） Positive expression rate of Ku70 in NPC group and nasal polyp group was 61.7%（29/47） and 30.0%（6/20） respectively,and the difference was of statistical significance（χ2=5.65,P〈0.05）.In different types of NPC issues,positive expression rate of Ku70 in groups of keratinizing squamous cell carcinoma and non-keratinizing carcinoma was 55.0% and 66.7% respectively（χ2=0.66,P〉0.05）.In different TNM NPC tissues,Ⅰstage ＋Ⅱstage group,and Ⅲ stage ＋Ⅳstage group,positive expression rate of Ku70 was 72.0% and 50.0% respectively,and the difference shows no statistic significance（χ2=2.40,P〉0.05）.The positive expression rate of Ku70 in NPC tissues with or without lymphatic metastasis is 73.3% and 35.5% respectively,and the differences had statistic significance（χ2=4.75,P〈0.05）.Conclusion It is suggested that Pokemon and Ku70 play important roles in NPC occurrence and development.Pokemon and Ku70 may become prognostic biomarker.
