摘要
目的:研究双点法注射6-羟基多巴胺(6-OHDA)制备的帕金森病(PD)大鼠相关分子的改变。方法:采用脑立体定位仪6-OHDA微量注射建立大鼠PD模型,2周后注射阿朴吗啡(APO),观察其行为学改变,并利用高效液相-电化学法测定正常大鼠(n=10)及PD大鼠(n=23)黑质组织中多巴胺(DA)及其代谢物3,4-二羟基苯乙酸(DOPAC)的含量;分光光度法测定超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量;RT-PCR法测定bcl-2和baxmRAN的表达。结果:与正常大鼠比较,PD大鼠黑质组织中DA及其代谢物DOPAC的含量降低(t=17.069和17.087,P均<0.001),SOD活性降低(t=6.314,P<0.001),MDA含量升高(t=11.060,P<0.001),bcl-2mRNA表达降低(t=9.333,P<0.001),而bax mRNA表达则升高(t=6.504,P<0.001)。结论:PD的发生与氧自由基损伤导致的神经元退变有关。
Aim:To investigate the changes of related molecules in Parkinson's disease(PD) rats induced by 6-hydroxydopamine(6-OHDA).Methods:PD models were established by microinjection of 6-OHDA solution into the substantia nigra of the right side of the brain.Then the behavior changes of rats were observed.The contents of DA and DOPAC were measured by HPLC-ED.SOD activity and MDA content were obtained by scenery spectrophotometry.The mRNA expressions of bcl-2 and bax were detected in substantia nigra by RT-PCR.Results:Compared with those of the normal rats,the levels of DOPAC and DA in substantia nigra of PD rats were decreased(t=17.069 and 17.087,P0.001),the SOD activity was lower(t=6.314,P0.001),the content of MDA was on the contrary(t=11.060,P0.001),the expression of bcl-2 decreased(t=9.333,P0.001),but the expression of bax increased in the PD rats(t=6.504,P0.001).Conclusion:PD may be related with oxygen free radicals and neuronal degeneration.
