摘要
[目的]获得小麦近等基因系TcLr24中NBS类抗病基因同源序列。[方法]利用已克隆植物抗病基因NBS序列中的保守结构"P-loop"和"GLPL"合成简并引物,以小麦近等基因系TcLr24基因组DNA为模板进行PCR扩增。[结果]得到13条具有连续ORF的抗病基因类似物序列,包括P-loop、Kinase-2、Kinase-3a、GLPL抗病基因所共有的保守结构,相应推测的氨基酸序列间的相似性系数在36.1%~98.3%,同时对分离的RGAs的氨基酸序列和5个已克隆植物NBS的氨基酸序列进行聚类分析表明,与Xa1、RPS2亲缘关系较近,而与Lr21亲缘关系较远。[结论]TcLr24中NBS类RGAs可能会有与Lr21不同机制的抗病功能。
[Objective] The study aimed to acquire the NBS-type resistance gene analogues in wheat TcLr24.[Method] Degenerate primers based on conserved motif(P-loop and GLPL)of the nucleotide binding site(NBS) region from the cloned plant disease resistance genes were used to isolate resistance gene analogues(RGAs) from genomic DNA of wheat TcLr24.[Result] 13 RGAs with uninterrupted open reading frames(ORFs) were obtained The analysis of RGAs amino acid sequence structures suggested that they contained the domains such as P-loop,Kinase-2,Kinase-3a,and GLPL,while the 13 RGAs deduced amino acid sequences showed identity ranged from 36.1% to 98.3%.Cluster analysis showed that 13 RGAs and RPS2 and Xa1 were clustered into one group.[Conclusion]Results showed that NBS type RGAs isolated from wheat TcLr24 might have different mechanism from Lr21.
