二甲肼诱发SD大鼠结直肠息肉模型探讨 被引量:3

摘要 目的建立一种简单、可行、稳定的结直肠息肉动物模型。方法 60只雄性SD大鼠随机分模型组(40只)和对照组(20只)。模型组皮下注射二甲肼(DMH)20mg.kg-1,对照组皮下注射与模型组DMH等体积的生理盐水,每周1次,持续18周。停药后继续喂养2周,自实验开始于第8、10、12、14、16及第18周分别处死模型组5只,对照组2只,其余大鼠全部于第20周处死。结果实验20周后,对照组无息肉生成,模型组共诱导出息肉83个,以增生性息肉和管状腺瘤为主;近、中及远段息肉之间相比较差异有统计学意义(P〈0.05),以中段和远段多发;与对照组比较,模型组大鼠体重下降明显,差异有统计学意义(P〈0.05);与第18~20周比较,第10~12周及14~16周诱导出的息肉数量减少,差异有统计学意义(P〈0.05)。结论二甲肼皮下注射可成功建立结直肠息肉SD大鼠模型。 Objective To establish a simple,practical,stable animal model of colorectal polyps.Methods Sixty male Sprague Dawley rats were randomly divided into two groups:the control group(n=20) and the model group(n=40).the model group were injected with 1,2-dimethydrazine(20mg/kg/week) and the control group were injected equivalent saline with model group.All animals received treatment eighteen weeks and continued feeding two weeks after stopping treatment.The rats were killed in batches at the 8th 、10th、12th、14th、16th and the 18th week of the experiment separately(two of the control group,five of the model).All remaining rats were killed at the 20th week.Results Twenty weeks later,the control group were non-polyp production,the model group had 83 polyps,which were primarily Hyperplastic polyp and Tubular adenomas;the number of Middle and distal polyps were more than that of Proximal(P0.05);the body weight of the rats in model group was lower than that in control group(P0.01);compared with the eighteenth to twentieth week,there was significant difference between the tenth to twelfth week and the fourteenth to sixteenth week in the number of polyps(P0.05).Conclusion The subcutaneous injection of DMH can establish colorectal polyps model in Sprague Dawley rat successfully and this model can be used for patients with polyps in colorectal cancer-related research.
