摘要
目的:观察 m I L12 基因转染的小鼠 E L4 细胞( C57 B L/6 小鼠 T 淋巴瘤细胞株)体外生物学特性的改变。方法:阳离子脂质体协同 I L12 重组腺病毒感染 E L4 细胞。结果及结论:基因转染的 E L4 细胞有 m I L12 的 m R N A 表达,培养上清中可以检测到 m I L12 的生物学活性。与野生型 E L4 细胞相比,m I L12 重组腺病毒感染的 E L4 细胞形态及体外增殖能力无明显改变, 转染细胞的成瘤性降低。此外,研究结果还表明,阳离子脂质体能显著提高腺病毒的感染效率。
Objective: To observe the biological characteristics of EL 4 cells transfected by murine interleukin 12 gene. Methods: Adenoviral vector containing mIL 12 was used to infect EL 4 cell line with the adjutant of lipofectamine. Results and Conclusion: mIL 12 Could be detected in the culture supernatant by ELISA. IL 12 mRNA Transcription could also be detected by RT PCR. In comparison with wild type EL 4 cells, mIL 12 modified EL 4 cells have no difference in growth. The tumorigenicity of mIL 12 modified cells was decreased. It was also found that lipofectamine could increase the efficiency of transfection by adenoviral vector.
