盐酸戊乙奎醚对感染性休克大鼠心肌核因子-κB的影响 被引量:2

摘要 目的探讨盐酸戊乙奎醚(长托宁)对感染性休克大鼠心肌的保护作用及其核因子-κB(NF-κB)机制。方法改良的盲肠结扎穿孔法(CLP)复制大鼠感染性休克模型。长托宁预防组(PP组):制作CLP模型,输注4mL/(kg.h)长托宁0.45mg/kg及生理盐水。长托宁治疗组(PT组):制作CLP模型输注生理盐水4mL/(kg.h),8h后改输注等量长托宁,然后给予等量生理盐水。多巴胺治疗组(DA组):制作CLP模型并输注生理盐水4mL/(kg.h),加入DA并调整用量。假手术组(C组)和休克组(S组)均给予等量生理盐水。术后监测心率(HR)和平均动脉压(MAP),取心肌组织测NF-κB含量并观察病理学改变。结果 CLP,6h后S组HR、MAP与C组、DA组、PP组比较,差异均有统计学意义(P<0.05);12h后S组与其余各组比较,差异均有统计学意义(P<0.01)。PP组、DA组和PT组NF-κB阳性细胞较S组显著减少(P<0.01)。PP组、DA组和PT组心肌损伤均较S组轻。结论长托宁能抑制感染性休克心肌NF-κB活化,预防性应用长托宁起到抗炎作用;治疗性应用长托宁可阻止器官功能衰竭,起到心肌保护作用。 【Objective】 To investigate the protection and mechanism of Penehyclindine Hydrochloride on myocardial NF-κ B in septic shock.【Methods】 Septic shock model was replicated by ligation and puncture(CLP).Group PP: established the model and 4 mL/(kg·h) of Penehyclidine Hydrochloride was injected with the concentration of 0.45 mg/kg.Then,saline was injected at the same speed.Group PT: established the model and injected saline with the speed of 4 mL/(kg·h).Penehyclidine Hydrochloride was changed after eight hours.Then,the saline was injected.Group DA: established the model,saline Added with dopamine was injected.Group C and S received saline only.After operation,HR and MAP were detected and the content of nuclear factor-κ B was measured.Also,the pathological changes of the myocardium should be observed.【Results】 Compared with group S,the change of HR and MAP was slight in other groups(P 0.01).The positive cell and average optical density value of NF-κ B was higher in group S than those in other groups(P 0.01).There were less myocardial injuries in group DA,PP and PT than those in group S.【Conclusion】 Penehyclindine Hydrochloride could inhibit the activtion of NF-κ B in septic shock and it could protect the myocardium.
