摘要
目的:探讨Hedgehog(Hh)信号通路对人胃癌SGC-7901细胞侵袭迁移的作用及其机制。方法:环靶明特异性阻断Hh信号通路后,Transwell小室和划痕实验检测SGC-7901细胞侵袭和迁移的能力;RT-PCR检测SGC-7901细胞中MMP-2和MMP-9 mRNA表达;细胞免疫化学检测SGC-7901细胞中MMP-2和MMP-9蛋白表达。结果:阻断Hh信号通路能显著抑制SGC-7901细胞的侵袭和迁移能力(P<0.05)。MMP-2和MMP-9的mRNA以及蛋白表达亦显著降低(P<0.05)。结论:Hh信号通路可能是通过调节MMP-2和MMP-9的表达促进SGC-7901细胞侵袭、迁移。
Objective:To investigate the effect and mechanism of Hedgehog signaling pathway on invasion and migration of human gastric cancer cell line SGC-7901.Methods: After cyclopamine blocked Hh signaling pathway specifically,transwell chamber assay and wound healing assay were used to examine the capacities of invasion and migration of SGC-7901 cells respectively.Reverse transcriptase-polymerase chain reaction(RT-PCR) was used to detect the expression of MMP-2 and MMP-9 mRNA in SGC-7901 cells.Immunochemistry was used to detect the expression of MMP-2 and MMP-9 protein in SGC-7901 cells.Results: The capacities of invasion and migration of SGC-7901 cells could be significantly inhibited after blocking Hh signaling pathway(P0.05).There was lower expression of MMP-2 and MMP-9 mRNA than the untreated group in SGC-7901 cells(P0.05).The expression of MMP-2 and MMP-9 protein also remarkably decreased(P0.05).Conclusion: Hedgehog signaling pathway may promote invasion and migration of SGC-7901 cells by regulating the expression of MMP-2 and MMP-9.
