2-氯环戊腺苷预处理延迟相对兔缺血-再灌注损伤心肌αB-晶状体蛋白的影响 被引量:1

摘要 目的研究腺苷A1受体激动药2-氯环戊腺苷(CCPA)预处理延迟相对缺血-再灌注损伤心肌αB-晶状体蛋白的影响。方法新西兰大白兔12只,随机均分为三组:假手术组(S组)、生理盐水预处理组(N组)、CCPA预处理组(C组)。S组分离兔左冠状动脉前降支,套带,但不阻断血流;N、C组在用药24h后分离兔左冠状动脉前降支,结扎30min,恢复灌注2h。提取心肌组织蛋白,采用WesternBlotting检测αB-晶状体蛋白的表达。结果 S组中αB-晶状体蛋白表达很少,C组αB-晶状体蛋白表达显著高于S、N组(P<0.01)。结论 CCPA预处理延迟性心肌保护作用与αB-晶状体蛋白表达上调有关。 Objective To investigate the effects of delayed preconditioning mediated by 2 chloro-N6-cyclopentyl-adenosine (CCPA) on myocardial αB crystallin. Methods Twelve rabbits were randomly divided into three groups: group S, group N and group C. CCPA was infused into rabbit in group C, while the same volume of normal saline(NS)was given instead in group N on day 1, no treatment was done in group S. 24 hours after CCPA or NS pretreatment, rabbits in groups C and N were subjected to 30 rain left anterior descending coronary artery(LAD) occlusion and reperfused for 2 h. Rabbits in group S did not subjecte to LAD occulsion and reperfusion. Myocardial αB crystallin was determinned with western blotting. Results αB crystallin was up-regulated by the CCPA pretreatment. Conclusion Delayed myocardial protection by CCPA pretreatment was relevant with upexpression of myocardial aB crystallin.
