摘要
目的探讨Src激酶在D1/D5受体激动剂诱导的脊髓长时程增强(LTP)中的作用。方法细胞外记录技术在脊髓腰膨大部记录背角浅层神经元C-纤维诱发电位。结果①D1/D5受体激动剂SKF38393(250μmol/L)诱导的脊髓LTP的效应可被D1/D5受体拮抗剂SCH23390(20μmol/L)所阻断;②Src激酶的选择性抑制剂Genistein(200μmol/L)对脊髓背角C-纤维诱发电位的基础电位没有影响,但可阻断SKF诱导的脊髓背角LTP;③N-甲基-D-天冬氨酸受体(NMDAR)亚基2B特异性拮抗剂Ro25-6981(50μmol/L)阻断SKF诱导的脊髓背角LTP。结论脊髓背角Src激酶和NMDAR2B受体参与D1/D5受体诱导的脊髓LTP。
Objective:To explore the role of Src kinase in the induction of spinal long-term potentiation(LTP) induced by the D1/D5 receptor agonist.Methods:C-fiber evoked field potentials were recorded at the superficial layer of the spinal dorsal horn at the lumbar enlargement by extracellular recording technique.Results:① SKF38393(SKF,250 μmol/L)-induced LTP of C-fiber evoked field potentials was blocked by SCH 23390(SCH,a D1/D5 receptor antagonist,20 μmol/L).② Genistein(200 μmol/L),a selective Src kinase inhibitor,blocked the induction of spinal LTP induced by SKF.③ NR2B antagonist Ro25-6981(Ro,50 μmol/L) prevented spinal LTP by SKF.Conclusion:Activation of Src kinase and NR2B in the spinal dorsal horn may be crucial for the induction of spinal LTP by D1/D5 receptor agonists.
