脊髓损伤后Wnt-3a表达对神经干细胞增殖的影响 被引量:10

摘要 目的研究大鼠脊髓损伤后Wnt-3a信号蛋白表达及其对脊髓神经干细胞增殖的影响。方法选用30只SD成年雌性大鼠,随机分为A组(正常对照组,n=5)和B组(脊髓损伤组,n=25)。A组不损伤脊髓,B组在大鼠T_(9~10)节段采用Allen打击法造成脊髓损伤,于造模后1、3、7、14、28 d取材。对距离损伤中心5 mm脊髓行免疫组化染色,检测和比较两组Wnt-3a表达动态变化及内源性神经干细胞增殖,并统计分析两者相关性。结果 A组大鼠脊髓中央管周围和外周软膜仅有极少量Wnt-3a表达,BrdU阳性细胞也不明显,白质中几乎没有。B组大鼠损伤1 d后脊髓中央管周围和外周软膜即有BrdU阳性细胞和Wnt-3a表达,3 d后表达明显增多,7 d后达到高峰,14 d后逐渐下降,28 d后仅有少量表达。统计软件分析显示Wnt-3a表达与Brdu表达具有相关性。结论大鼠脊髓损伤后Wnt-3a表达增多,可能促进内源性神经干细胞增殖。 Objective To investigate the effect of Wnt-3a on the proliferation of neural stem cells after spinal cord injury in rats.Methods Thirty Sprague-dawley(SD) female rats were randomly divided into two groups:the control group(group A,n = 5) and the injury group(group B,n = 25).The spinal cords of group A were not injured and the spinal cords of group B were injured by the Allen method.The spinal cord of models was took 1d,3 d,7 d,28 d after injury and stained them with method of immunohistochemistry.The expression of Wnt-3a was tested and the relativity between Wnt-3a and the proliferation of neural stem cells was analysed.Results We found that at the central canal and the outer circumference of the spinal cord of group A, there were little Wnt-3a proteins and BrdU positive cells.But at the central canal and the outer circumference of the spinal cord of group B,there were some Wnt-3a proteins and BrdU positive cells 1 day after injury and the expression increased obviously 3 days after injury.They reached the highest point or level 7 days after injury and declined gradually 14 days after injury,and leaved a little Wnt-3a proteins and BrdU positive cells 28 days after injury.There were positive relations between the expression of Wnt-3a proteins and BrdU positive cells though the analyse of statistic.Conclusion The expression of Wnt-3a proteins are enhanced after injury,which probably facilitate the proliferation of neural stem cells.
