摘要
目的建立测定人工养殖蟒蛇胆汁中牛磺蟒胆酸钠含量的方法。方法采用高效液相色谱法测定其含量。色谱条件:Phenomenex Luna C18色谱柱(250 mm×4.60 mm,5μm);流动相为乙腈-0.15%磷酸氢二钠(10%磷酸调pH至3.0),梯度洗脱;流速为1.0 mL.min-1;检测波长203 nm;柱温35℃;进样量10μL。结果牛磺蟒胆酸钠在20.2~322.4μg.mL-1(r=0.998 9)浓度范围内线性关系良好,平均回收率为102.4%(RSD=1.4%,n=6);8批蟒蛇胆汁中牛磺蟒胆酸钠含量差异较大,平均含量为65.09 mg/g-1。结论本方法准确、快速、灵敏、重复性好,可用于蟒蛇胆汁的质量控制。
Objective To establish a method for the determination of sodium tauropythocholate in the bile of artificial cultured python.Methods High performance liquid chromatography(HPLC) was carried out on a Phenomenex Luna C18 column(250 mm × 4.60 mm,5 μm) with the mobile phase consisting of acetonitrile and 0.15 % disodium hydrogen phosphate(adding 10 % phosphoric acid to adjust pH value being 3.0) and by using a linear gradient elution.The flow rate was 1.0 mL·min-1,and UV detection wavelength was set at 203 nm.The column temperature was 35 ℃ and the injection volume was 10 μL.Results The calibration curve of sodium tauropythocholate was linear in the concentration range of 20.2 ~ 322.4 μg·mL-1(r=0.998 9),and the average recovery rate was 102.4 %(RSD=1.4 %,n=6).There was great difference of sodium tauropythocholate content in 8 batches of python bile,and the average content was 65.09 mg/g.Conclusion The established method is precise,rapid and accurate,which can be used for quality control of python bile.
