Immunopathologic studies on the effects of immunosuppressant leflunomide on penetrating keratoplasty rejection in rats model

摘要 目的:研究角膜移植排斥反应的免疫病理学变化,阐明Leflunomide的免疫抑制机理。方法:应用免疫组织化学染色方法检测环孢霉素A、Leflunomide及对照组角膜移植片中CD4+细胞、CD8+细胞、白细胞介素-2受体及主要组织相容性Ⅱ类抗原的表达。结果:排斥的角膜组织大量表达上述免疫细胞和分子,Leflunomide可以对这些细胞和分子产生明显的抑制作用。结论:迟发型超敏反应和细胞毒性T细胞的作用与植片的免疫性损伤密切相关,白细胞介素-2受体和主要组织相容性Ⅱ类抗原的异常表达在排斥反应中具有重要的作用,Leflunomide能抑制淋巴细胞的活化、增殖,延长角膜移植物的存活时间。 To study the immunopathologic process of rejection and elucidate the immunosuppressive mechanism of leflunomide By using immunohistochemical techniques,the expression of CD4 + cells,CD8 + cells,IL-2 receptor,MHC-Ⅱ antigen in rejected corneal grafts were studied Immunohistochemical studies showed diffuse infiltration of CD4 + cells,CD8 + cells and strong expression of IL-2 receptor and MHC- Ⅱ antigen in rejected corneal grafts,especially in the acute rejection stage Leflunomide can inhibit the expression of the cells and molecules in the grafts Keratoplasty rejection is significantly related to delayed type of hypersensitivity and cytotoxic T cells,and IL-2 receptor and MHC-Ⅱ antigen also affect the rejection process;Leflunomide prolongs the survival time of corneal allografts by inhibiting T cell activation and proliferation
