不同干预方法对肥胖大鼠血清FFA、TNFα-的影响 被引量:2

Different intervention effects on serum FFA and TNF-α levels of high-fat-diet fed obesity rats

摘要 目的探讨肥胖对大鼠血清游离脂肪酸(FFA)、肿瘤坏死因子-α(TNF-α)的影响,并观察二甲双胍及游泳运动对肥胖大鼠血清FFA、TNF-α的影响。方法 SD大鼠70只随机分为正常饲养(NC)组和高脂饲养(HF)组,16周后HF组成功建立肥胖模型后,再随机分为:肥胖对照(OB)组、二甲双胍(MET)组、游泳运动(SWI)组、二甲双胍+游泳运动(MAS)组。检测干预前及干预后各组体重(BW)、空腹血糖(FPG)、甘油三酯(TG)、总胆固醇(TCH)、极低密度脂蛋白胆固醇(VLDL-C)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、血清FFA和TNF-α。结果与NC组比较,HF组大鼠BW、FPG、TG、TCH、VLDL-C、FINS、HOMA-IR、血清FFA和TNF-α水平均显著增高(P<0.05);与OB组比较,MET、SWI和MAS组大鼠BW、HOMA-IR、FFA、TNF-α均显著降低(P<0.05);大鼠血清FFA水平以及TNF-α水平与HOMA-IR呈正相关(P<0.05),血清FFA水平与TNF-α水平呈正相关(P<0.05)。结论二甲双胍和游泳运动可显著降低肥胖大鼠血清FFA、TNF-α水平,减轻肥胖大鼠机体炎症状态,改善胰岛素抵抗,血清FFA与TNF-α对胰岛素抵抗可能有协同作用。 Objective To explore the effects of obesity on the FFA,TNF-α level of rats and investigate the effects of metformin,swimming exercise on FFA,TNF-α levels of high-fat-fed obesity rats Methods 70 SD rats were randomly divided into two groups in which 10 rats were fed with normal diet as normal control group(NC),and the other 60 were fed with high fat diet as high fat diet group(HF).After 16 weeks obesity model was established from HF group and was randomly divided into four groups:obesity group(OB),metformin group(MET),swimming exercise group(SWI),and the combination of metformin and swimming exercise group(MAS) with 10 rats in each group.Body weight(BW),fasting plasma glucose(FPG),triglyceride(TG),total cholesterol(TCH),fasting insulin(FINS),HOMA-IR and fasting FFA and TNF-α were detected before and after treatment.Results Compared with NC group,BW,FPG,TG,TCH,HOMA-IR,FINS and FFA,TNF-α levels of each groups in obesity model increased significantly(P0.05).BW,HOMA-IR,FFA,TNF-α in MET group,SWI group and MAS group were down-regulated compared to those in OB group(P0.05).Correlation analysis found that the level of TNF-α correlated with the level of HOMA-IR.The levels of FFA and TNF-α were correlated with the level of HOMA-IR.The level of FFA was correlated with the level of TNF-α.Conclusion Metformin and swimming exercise can significantly reduce the level of serum FFA and TNF-α,the inflammatory status of obese rats,and can improve insulin resistance.The serum FFA and TNF-α on insulin resistance may be synergistic.
