摘要 目的探讨环氧合酶-α抑制剂NS-398对人胰腺癌细胞株PCNA-1的增殖抑制作用。方法采用MTT法、流式细胞术检测NS-398对胰腺癌细胞株PCNA-1细胞增殖抑制、细胞周期及对Bcl-2蛋白表达的影响。结果 NS-398抑制胰腺癌细胞株PCNA-1的增殖活性,呈剂量依赖效应关系;NS-398可影响PCNA-1细胞周期变化;细胞经80μmol/L的NS-398处理48 h后bcl-2蛋白与对照组相比表达下调(P<0.01)。结论 NS-398对胰腺癌细胞株PCNA-1有抑制增殖、诱导其凋亡作用,诱导细胞凋亡的机制可能与细胞凋亡相关基因bcl-2表达下调有关。 Objective To investigate the effects of cyclooxygenase inhibitor NS-398 on the proliferation and apoptosis of PCNA-1 and its influence of bcl-2 protein.Methods The effects of NS-398 on the proliferation of human PCNA-1was detected by MTT assay,The effects of NS-398 on the cell cycle of human PCNA-1 and the expression of apoptosis associated protein bcl-2 were detected by flow cytometry.Results NS-398 inhibit the proliferation of human PCNA-1 as dose-dependent effect,NS-398 can influence the cell cycle;dealt with 80umol/L NS-398 for 48h,the expression of bcl-2 was down regulation compared with control group(P0.01).Conclusion NS-398 can inhibit the proliferation and induce apoptosis of PCNA-1,the mechanism of apoptosis is related to the down regulation of bcl-2.
