摘要
探索从苦碟子中提取总黄酮的最佳工艺。采用分光光度法测定总黄酮含量,通过单因素试验分析了乙醇浓度、回流时间、提取次数、溶剂用量4个主要因素对苦碟子总黄酮提取率的影响,并在单因素试验的基础上通过正交设计法优化了苦碟子总黄酮提取工艺条件。影响苦碟子总黄酮的主次因素为:乙醇浓度>回流时间>溶剂用量>提取次数。苦碟子总黄酮最佳提取工艺为:10倍量60%乙醇回流提取2次,每次2h。
To study and optimize the extraction procedure for total flavonoids from Ixeris Sonchifolia (Bge.) Hance. Spectrophotometry was adopted to determine content of total flavonoids in Ixeris Sonchifolia (Bge.) Hance. Effects of alcohol concentration, extraction duration, extraction times, volume of alcohol on extraction rate of total flavonoids were analyzed using single-factor test. Based on this, the extraction technological conditions of total flavonoids from Ixeris Sonchifolia (Bge.) Hance were optimized through orthogonal design method. The factors that influenced the extraction rate were sorted as follows: the concentration of ethanol〉extraction time〉volume of ethanol 〉heated times. The optimal process of the extraction technology which obtained through orthogonal design is: 60 % ethanol, ten-fold solvent, two times extraction, two hours per extraction time.
