摘要
目的探讨黄芪甲苷对脐带血间充质干细胞向心肌细胞分化的作用。方法采用贴壁法分离人脐带血间充质干细胞,用流式细胞术对细胞表型进行鉴定。通过免疫组化方法观察黄芪甲苷对间充质干细胞向心肌细胞分化的促进作用。结果流式细胞仪检测结果表明,分离的脐带血间充质干细胞表达CD29、CD44和CD105,不表达CD34和HLA-DR。黄芪甲苷(浓度200 mg/L)作用下,间充质干细胞表达特异性心肌细胞蛋白,包括结蛋白(desmin)、α横纹肌肌动蛋白(-αsarcomeric actin)和肌钙蛋白T(C-TnT)。结论黄芪甲苷对脐带血间充质干细胞分化的促进作用,分化的心肌细胞在心肌损伤性疾病的潜在的治疗价值值得进一步研究。
Objective To evaluate whether Astragaloside could promote the differentiation of cord blood-derived mesenchymal stem cells toward cardiomyocytes.Methods Mesenchymal stem cells were isolated from cord blood by repeated plating.The phenotypes of stem cells were assayed by FACS.Differentiation of mesenchymal stem cells was shown by immunochemistry after stem cells were cultured in the presence of Astragaloside.Results FACS showed that cord blood-derived mesenchymal stem cells expressed high levels of CD29,CD44 and CD105,but were negative for CD34 and HLA-DR.Astragaloside(200mg/L) could promote the differentiation of cord blood-derived mesenchymal stem cells toward cardiomyocytes,which was indicated by expression of desmin,α-sarcomeric actin and C-TnT after culture.Conclusion Astragaloside could promote the differentiation of cord blood-derived mesenchymal stem cells toward cardiomyocytes and its therapeutical potential for heart disease disserves further study.
