摘要 目的比较慢病毒载体和腺病毒载体转染人脐带血间充质干细胞后,外源基因的表达。方法采用人脐带血分离培养间充质干细胞,制备表达GFP的慢病毒载体和腺病毒载体,用以转染间充质干细胞,观察其外源基因的表达。结果脐带血间充质干细胞与骨髓中的类似,其外表呈纤维样;转染5周后,慢病毒载体转染的GFP阳性表达明显高于腺病毒载体。结论慢病毒载体较腺病毒载体作为基因转导系统更有优势。 Objective To compare between expression of exogenous genes in umbilical blood-derived mesenchyrnal stem cells, which were transfected by lentivirus vectors and adenovirus vectors. Methods Mesenchymal stem cells were isolated and from human umibilical cord blood. Lentivirus vectors and adenovirus vectors expressing GFP were constructed, then mesenchymal stem cells were transfected to observe the expressions of exogenous genes. Results Umbilical blood-derived mesenchyrnal stem cells were similar to the cells derived from bone marrow, they were fibroblast-like cells. Five weeks later, positive expression of GFP which was transfected by lentivirus vectors was obvious superior to that of adnovirus vectors. Conclusions Lentivirus vectors have more advantages as a gene transferring (gene conduction) system than adenovirus vectors.
