摘要
目的研究大肠癌组织的细胞周期分布状态与临床病理因素的关系,以及和细胞周期调节蛋白的表达水平是否相关。方法应用流式细胞术检测45例大肠癌临床标本细胞周期中G0/G1、S、G2/M期所占的比例,并检测每例标本的细胞周期蛋白CyclinD1、CyclinE、CyclinA、CyclinB1的表达水平。结合临床病理因素进行统计分析。结果肿瘤细胞G0/G1、S、G2/M期所占的比例和患者性别、年龄、肿瘤部位、病理分级、浸润深度、淋巴结转移、Dukes分期均无明显相关性,均P>0.05。G0/G1期的比例与CyclinD1、CyclinE,S期与CyclinE、CyclinA,G2/M期与CyclinA的表达水平均不相关,均P>0.05;但是G2/M期的比例与CyclinB1的表达水平正相关,r=0.335,P=0.035。结论大肠癌肿瘤细胞的周期分布和临床病理因素无关,肿瘤细胞周期蛋白的表达特点与体外培养的细胞系不同,表现得更加复杂多样。
Objective To investigate the association between cell cycle status and clinicopathological characteristics or cyclins levels of colorectal cancer.Methods DNA profiles of 45 cases of colorectal cancer were detected by flow cytometry.Populations of G0/G1,S and G2/M phases were assessed.The expression of cyclinD1,cyclinE,cyclinA,and cyclinB1 was also detected by flow cytometry.Results Proportions of G0/G1,S,and G2/M phase in tumor tissues were not associated with patients' age,sex,tumor differentiation,lymphatic invasion,depth of tumor invasion and Dukes stage.CyclinD1 and cyclinE levels were not correlated with proportion of cells in G0/G1phase,cyclinE and cyclinA levels not with that in S phase,and cyclinA level not with that in G2/M phase,but there was a positive relationship between cyclinB1 level and proportion of cells in G2/M phase.The proportion of cells in G2/M phase was higher in tumors with high cyclinB1 expression.Conclusion The cell cycle distribution of colorectal cancer cells isolated from clinical specimens was not associated with clinicopathological characteristics.The expression patterns of cyclins were diverse,different from those of tumor cell lines in vitro.
