摘要
目的 探讨米托蒽醌治疗白血病的机制。方法 应用细胞培养,细胞形态学观察,DNA琼脂糖凝胶电泳等技术进行检测和观察。结果 HL-60白血病细胞在米托蒽醌的作用下,出现典型的凋亡特征,细胞形态学表现出细胞核裂解,染色质聚集,核碎裂,胞浆浓缩,有空泡形成,DNA琼脂糖电泳出现典型梯形条纹。结论 米托蒽醌通过诱导白血病细胞凋亡起到治疗白血病的作用。
To investigate the mechanisms of mitoxantrone killing leukemia cells. Methods Culture HL 60 cells, cell morphollogy, DNA agarose gel electrophoresise were performed. Results HL 60 leukemia cells treated with mitoxantrone underwent apoptosis. Apoptosis cells demonstrated compaction and segregation of the nuclear chromatin, condensation and vacuolus of the cytoplasm. Nuclear DNA of apoptosis cells display ladder bands charateristic of internucleosomal DNA fragmentation. Conclusion The mechanism of mitoxantrone killing leukemia cells is inducing apoptos is. Key Words mitoxantrone; leukemia; apoptosisis medicines,abuse of antibiotics and incorrect conception of consumption.It is suggested that the effective measures to control the over-consuming of medicines in our country are to adhere to the policy of the development of non-patent medicines and the practice of unified purchase and marketing of medicines,cut off unfair competition,and illegal way of selling medicines,work out a name list of basic medicines,attach great importance to economical evaluation,carry out standardized medical care,control hospital infection,change or correct the conception of consumption.
