Advances in Expression of Recombinant Protein in E.Coli Periplasmic Space

摘要 大肠杆菌表达系统是基因表达技术中发展最早和应用最广的表达系统,由于大肠杆菌周质空间具有其独特优势,重组蛋白在周质表达和提取技术得到国内外学者的关注。文章从重组蛋白在大肠杆菌周质表达的特点、提高周质蛋白表达的策略以及周质蛋白提取技术进行了论述。 Cloning and expression of recombinant protein in Escherichia coli has been developed for a long time and it is widely used as a classical expression system now.Because of the unique advantages in E.coli periplasmic space,the expression and extraction of periplasmic protein have received the attention by the scholars both at home and abroad.The characteristics of expression of recombinant protein in E.coli,the strategies for high-level expression and the release technology were reviewed in this article.
