摘要
建立了HPLC法测定微乳中DHA含量的方法。采用无水硫酸铵对微乳进行破乳处理,对DHA藻油进行皂化等处理,采用优化的DHA测定条件为:以C18色谱柱,流动相为乙腈:四氢呋喃:水(含0.4%HAC)=77:3:20,柱温30℃,流速1.0 mL/min,紫外检测器检测,波长215 nm。结果显示,DHA在14.06~449.92μg/mL内呈良好的线性关系(R2=0.9997),平均回收率为101.56%,RSD=2.11%(n=6)。实验结果表明该方法可靠、简便、重现性好,可用于微乳中DHA含量测定。
The method of detecting the contents of DHA in microemulsion by HPLC was developed.After breaking of the emulsion with ammonium sulfate treatment,DHA algal oil was treated by saponification,The optimized condition was: the sample was analyzed in a RP-C18 column with acetonitrile tetrahydrofuran water(containing 0.4% HAC)=77:3:20 as the mobile phase,and the UV detector was used in HPLC.The wave length was 215 nm.Column temperature was fixed at 30 ℃ and the flow rate was 1.0 mL/min.The results show that the standard curves of DHA presented the favorable linear correlation over the range of 14.06~449.92 |ìg/mL.The average recovery rate of DHA are 101.56%,and the RSD is 2.11%(n=6).The results show that the present method is reliable and relatively simple with a good reproducibility for the determination of DHA in microemulsion.It can be used as method of detection of the determination of DHA in microemulsion.
