人蚤实验种群的建立和保持以及遗传稳定性的监测 被引量:1

ESTABLISHMENT AND MAINTENANCE OF LABORATORY POPULATION OF PUBLES IRRITANS AND MONITORING FOR ITS GENETIC VARIATION

摘要 在实验室建立了人蚤实验种群,运用了交叉传代法以保持种群的遗传稳定性;同时运用RAPD技术对人蚤实验种群遗传结构的时间变化作了监测,以确认遗传结构的稳定和传代方法的有效性。结果表明,在5个的时间内,人蚤实验种群的遗传结构没有发生大的变化,种群内遗传距离分别为0.2088±0.0517和0.2140±0.0787。说明交叉传代法对保持实验种群的遗传稳定性是有效的,RAPD技术在遗传稳定性监测方面的运用也是可行的。同时对RAPD技术在这方面运用时的一些问题作了讨论。 The Random Amplified Polymorphic DNA PCR was used to monitor the temporal genetic variation of laboratory population of Pulex irritans,by which the author studied the population dynamics of nature population.The main purpose of this study was to ascertain whether the genetic construction of the population changed when maintained in laboratory.The results showed little temporal genetic change in 5 molnths(0208800517 and 0.21400.0787 Nie's genetic distance,respectively),and the method to maintain the population could be accepted.In addition,the property of RAPD that can affect its using in temporal genetic variation monitoring is discussed.
