当归补血汤载药血清对骨髓基质细胞增殖及细胞因子表达的影响 被引量:16

Effect of Serum Containing Drug of Danggui Buxue Decoction on the Proliferation of Mice Bone Marrow Stromal Cells and Expression of Cytokines

摘要 目的:观察当归补血汤载药血清对小鼠骨髓基质细胞增殖及细胞因子分泌的影响。方法:分离骨髓基质细胞,接种于96孔板培养,在相差显微镜下观察细胞形态和生长状况;将细胞分4组,用含有不同剂量的当归补血汤载药血清作用,采用MTT法检测各组细胞的增殖活性及ELISA法检测造血生长因子GM-CSF和IL-3的表达情况。结果:在不同浓度的载药血清作用下,骨髓基质细胞增殖和分泌GM-CSF和IL-3显著(P<0.05),呈剂量依赖性,但浓度过高可减弱细胞的增殖分泌效应。结论:当归补血汤载药血清能够促进骨髓基质细胞的增殖以及GM-CSF和IL-3的分泌。 Objective:To investigate the effect of serum containing drug of Danggui Buxue Decoction on the proliferation of mice bone marrow stromal cell(MBSCs) and expression of cytokines.Methods:MBSCs of the new born mouse were procured.MBSCs were cultured in the 96 holes board by using the method of serum-free incubation technique and the growth characteristic of cells was observed.MBSCs were divided into 4 groups and then MBSCS were cultured at different concentrations in the serum containing drug of Danggui Buxue Decoction.MTT method was used to observe the effects of various culture fluid on proliferation of MBSCs and ELISA method was used to observe the expression level of hematopoietic growth factors GM-CSF and IL-3.Results:After the stimulation of various culture fluid,the results suggested MBSCs showed a proliferative property manifesting a higher cell number.The expression level of GM-CSF and IL-3 of each serum containing drug of Danggui Buxue Decoction group was significantly higher than the normal control.All had significant difference.But with heightener of the concentration of Danggui Buxue Decoction the proliferation of MBSCs and the expression level of GM-CSF and IL-3 were weakened.Conclusion:The serum containing drug of Danggui Buxue Decoction promote the proliferation of MBSCs significantly and the expression level of GM-CSF and IL-3.
