子宫颈癌组织中COX-2与VEGF的表达及相关性研究 被引量:2

The study on expressions and correlationship of COX-2 and VEGF in cervical cancer

摘要 目的:研究环氧合酶-2(Cyclooxygenase,COX-2)及血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)在子宫颈癌及宫颈上皮内瘤样病变Ⅱ、Ⅲ组织中的表达,以探讨COX-2的表达与子宫颈癌发生、发展的关系以及与VEGF之间的相关性。方法:应用逆转录聚合酶链反应(RT-PCR)技术,检测42例子宫颈癌,10例宫颈上皮内瘤样病变Ⅱ、Ⅲ,10例正常宫颈中COX-2 mRNA和VEGF mRNA的表达。结果:COX-2 mRNA,VEGF mRNA在正常、宫颈上皮内瘤样病变Ⅱ、Ⅲ组织及子宫颈癌中的相对含量分别为8.86±19.12、55.76±48.46、132.64±56.99和15.23±24.86、57.30±49.40、142.31±59.45。子宫颈癌及上皮内瘤样病变Ⅱ、Ⅲ组织中COX-2 mRNA,VEGF mRNA的相对含量显著高于正常宫颈组织,差异有统计学意义(P<0.01),子宫颈癌COX-2 mRNA,VEGF mRNA的相对含量高于上皮内瘤样病变Ⅱ、Ⅲ组织,差异有统计学意义(P<0.01)。且COX-2的表达与VEGF的表达呈正相关(r=0.959,P<0.01)。结论:COX-2、VEGF在子宫颈癌组织中高表达,与其发生、发展有密切的关系,VEGF可能是COX-2促进子宫颈癌形成、发展的主要机制之一。 Objective: To investigate the relationship between the expressions of COX -2 mRNA and VEGF mRNA in carcinogenesis and development of the cervical cancer. Methods: Quantitive reverse transcription -polymerase chain reaction (RT -PCR) was used to examine the levels of COX -2 mRNA and VEGF mRNA in the following groups: 10 cases with normal cervical tissue , 10 cases with atypical hyperplasia cervical tissue and 42 cases with cervical'cancer. Results: The relative content of COX -2 mRNA and VEGF mRNA in normal cervical tissue, atypical hyperplasia cervical tissue and cervical cancer was 8.86 ±19. 12, 55.76±48.46, 132. 64 ±56. 99 and 15.23 ±24. 86, 57. 30 ± 49. 40, 142. 31 ±59. 45 respectively, and the expressions in cervical cancer and atypical hyperplasia cervical tissue were significantly higher than that in normal cervical tissue ( P 〈 0. 01 ). The relative content of COX - 2 mRNA and VEGF mRNA in cervical cancer was greater significantly than that in atypical hyperplasia cervical tissue ( P 〈 0. 01 ) . COX - 2 was positively correlated with VEGF. Conclusion: High expressions of COX - 2 and VEGF are detected in cervical cancer tissue. It plays an essential role in tumor development and growth. COX -2 may stimulate angiogenesis through VEGF up -regulation.
